摘要: Fiber_FISH(fiber based fluorescent in situ hybridization DNA纤维荧光原位杂交)是近年来发展起来的一项直接在DNA纤维上进行荧光原位杂交的技术,其分辨率和灵敏度可分别达到1~2 kb和200 bp。描述了Fiber_FISH的技术要点,念珠状信号形成的可能原因,以及这一技术在植物基因组研究中的应用。

Abstract: Fiber_FISH is a technique recently developed to detect directly DNA sequences on extended DNA fibers by fluorescence in situ hybridization (FISH), the resolution and sensitivity of which are improved to 1~2 kb and 200 bp respectively. This paper describes the key points of Fiber_FISH technique, the probable nature of the beaded fluorescent spots and the application of Fiber_FISH technique in plants genome research.
