摘要: 泛肽(Ubiquitin 简称Ub)是一个由76个氨基酸残基组成的非常保守的小蛋白质。泛肽依赖性的蛋白质降解途径(Ubiquitin-dependebt proteiytic pathway)是目前已知的最重要的、有高度选择性的蛋白质降解途径。泛肽系统由Ub、Ub活比酶、Ub结合酶、Ub-蛋白质连接酶、Ub-C末端水解酶和26S蛋白酶体组成。本文详细地介绍了泛肽系统各个组成部分的种类、结构与功能,蛋白质泛肽化及其降解机制和底物识别模式。

Abstract: Ubiquitin (Ub) is a conserved small protein consisted of 76 amino acid residues. Ubiquitin-dependent proteolytic pathway is the most important selective proteolytic pathway. Ubiquitin system is composed of ubiquitin, ubiquitin-activating enzymes, Ubiquitin-conjugating enzymes, ubiquitin-protein ligases, ubiquitin-C-terminal hydrolase and 26S proteasomes. This review discussed and described components of ubiquitin system, structure and function of each component, machanisms of protein ubiquitination and degradation of ubiquitinated proteins, and modes of recognition of substrates.
