三种瓶尔小草科植物的比较代谢组学研究

Comparative metabolomics study of three Ophioglossaceae species

摘要: 瓶尔小草科(Ophioglossaceae)为多年生草本蕨类植物,其野生资源种类丰富,兼具食用与药用价值。该科植物常以全草入药,具有清热解毒的功效,临床用于肺热咳嗽、毒蛇咬伤等病症。《中华本草》收载多种瓶尔小草科植物,其中以瓶尔小草(Ophioglossum vulgatum)与阴地蕨(Sceptridium ternatum)最常见。化学成分研究表明,瓶尔小草与阴地蕨均含有类黄酮、萜类、酚酸类等生物活性成分。然而,目前该科植物中已鉴定的化学成分仍较为有限,制约了其药用价值的深入开发与利用。本研究采用广泛靶向代谢组学技术,对瓶尔小草、心叶瓶尔小草(O. reticulatum)及阴地蕨的全草样本进行系统分析,比较三者代谢物组成差异。共鉴定出2651种代谢物,主要包括氨基酸及其衍生物、脂质、有机酸、核苷酸及其衍生物等初级代谢物,以及类黄酮、生物碱、萜类、酚酸、木脂素与香豆素等次生代谢物。次生代谢物种类多于初级代谢物,其中类黄酮化合物数量最丰富,以黄酮醇和黄酮为主。代谢谱比较分析表明,同属的瓶尔小草与心叶瓶尔小草在代谢物组成上更为接近,且二者所含类黄酮物质均显著多于阴地蕨,反映出代谢物组成与物种分类地位之间的相关性及种间特异性。三种植物中,瓶尔小草所富集的代谢物种类最多,阴地蕨次之,心叶瓶尔小草最少。进一步比较不同采收期瓶尔小草代谢物积累动态,发现八月采集样本中类黄酮与萜类含量最高。本研究系统揭示了三种瓶尔小草科植物代谢物的多样性、种属间差异以及采收期对代谢物积累的影响,为其后续药用开发与营养潜力评价提供了科学依据。

Abstract: Ophioglossaceae species are perennial herbaceous ferns with rich wild diversity, exhibiting significant edible and medicinal value. Plants of this family are commonly used in their entirety in traditional medicine for their heat-clearing and detoxifying effects, and are clinically applied in treating conditions such as lung heat cough and snakebites. Chinese Materia Medica includes several species from this family, among which Ophioglossum vulgatum and Sceptridium ternatum are the most common. Phytochemical studies have shown that both O. vulgatum and S. ternatum contain bioactive compounds such as flavonoids, terpenoids, and phenolic acids. However, the number of chemically characterized constituents in this family remains limited, which restricts further exploration of their medicinal potential. In this study, a widely targeted metabolomics approach was applied to systematically analyze the metabolic profiles of whole plants of O. vulgatum, O. reticulatum, and S. ternatum. A total of 2,651 metabolites were identified, including primary metabolites such as amino acids and their derivatives, lipids, organic acids, nucleotides and derivatives, as well as secondary metabolites such as flavonoids, alkaloids, terpenoids, phenolic acids, lignans, and coumarins. The diversity of secondary metabolites exceeded that of primary metabolites, with flavonoids being the most abundant, predominantly flavonols and flavones. Comparative metabolomic analysis revealed that the two congeneric species, O. vulgatum and O. reticulatum, exhibited closer metabolic profiles, and both contained significantly more flavonoids than S. ternatum, indicating phylogenetic consistency and interspecific specificity in metabolite composition. Among the three species, O. vulgatum showed the highest number of enriched metabolites, followed by S. ternatum, and O. reticulatum had the fewest. Furthermore, analysis of O. vulgatum samples collected at different harvesting periods indicated that samples collected in August exhibited the highest accumulation of flavonoids and terpenoids. This study systematically elucidates the diversity of metabolites in three Ophioglossaceae species, the metabolic variations among species and genera, and the influence of harvesting time on metabolite accumulation, thereby providing a scientific basis for future medicinal development and nutritional evaluation.
