A Rapid Micro-volume Method for Determining b-amylase Activity in Plant Seeds

摘要: 针对3, 5-二硝基水杨酸(DNS)法的缺点, 以对硝基苯酚麦芽五糖(PNPG5)为底物, 建立并改进了植物种子b-淀粉酶活性测定的方法。本方法具有微量和快速的特点; 进一步通过对4种植物材料种子的酶活性测定以及a-淀粉酶的干扰评估实验, 证实了该法可以运用于植物种子b-淀粉酶活性的专一性测定。

Abstract: In view of the disadvantages of the 3,5-dinitrosalicylic acid (DNS) method, we developed a rapid micro-volume procedure utilizing p-nitrophenyl-a-maltopentaoside (PNPG5) as a substrate for specific determination of b-amylase activity. The assay procedure was evaluated with seeds from 4 plant species, and the exact degree of interference by a-amylase was determined. This modified method could be applied to the selective measurement of b-amylase activity in different plant seeds.
