In vitro Regeneration of Achnatherum splendens (Trin.) Nevski

Abstract: We studied the regeneration of Achnatherum splendens (Trin.) Nevski in vitro and selected several favorable media.Germ-free plants were produced with germination on MS medium. Callus was induced with B5+1.5 mg.L-1 2,4-D+0.5 mg.L-1 NAA,with leaf sheath and hypocotyls as explants. After two subcultures, shoots and roots were induced. The medium containing B5+0.5 mg.L-1 6-BA + 0.2 mg.L-1 NAA was suitable for shoot induction. For root induction, the medium containing 1/4 B5+1.0 mg.L-1 NAA+ 0.2 mg.L-1 IBA +1.0 g.L-1 active carbon was favorable. With this regeneration system for A. splendens (Trin.) Nevski, we achieved the highest survival rate of transplants.
