A Method to Improve Sample Preparation for Observing Stomata Guard Cells in Triticum aestivum

Abstract: We developed an efficient whole-leaf clearing method for observing stomata guard cells of anther-derived plants in Triticum aestivum. We compared the effect of 4 different clearing agents. With this method, we could obtain a sample for observing stomata guard cells in wheat without mesophyll tearing and shaving. After leaf treatment with glycerite, saturation with chloral hydrate and mixed solution of chloral hydrate and glycerin revealed clear images of stomata guard cells by microscopy.
