A Regeneration System and Ploidy Identification of Plantlets for Endosperm of Dioscorea zingiberensis

Abstract: We studied the effect of various plant growth regulators on inducing endosperm callus and plant regeneration using the endosperm of Dioscorea zingiberensis as explants and identified the ploidy of plantlets. The optimal medium for inducing callus, adventitious bud differentiation and root formation was MS+2.0 mg·L–12,4-D+0.5 mg·L–16-BA, MS+2.0 mg·L–16-BA+0.1 mg·L–1NAA and 1/2MS+0.3 mg·L–1NAA, respectively. On observing the growth points of axillary buds in regenerated plants by chromosome slice production techniques, 10%, 16% and 74% of total observed cells had 20, 21–29 and 30 chromosomes, respectively. We obtained triploid plantlets.
