Rapid, Effective Method for Anthocyanin Analysis in Tobacco Corolla

Abstract: The analysis of anthocyanins is an important task for studies of flower coloration. This paper describes a rapid, effective method for analysis of anthocyanin composition and content in tobacco corolla based on high-performance liquid chromatography-electrospray ionization-mass spectrometry. HPLC analysis time was shortened to 15 min by this method, and the separation effect was guaranteed. The analysis speed is largely improved, and costs are reduced. The analysis of anthocyanins in transgenic tobacco was successful with this method, so it can be used in a general laboratory, even without advanced instruments such as ultra performance liquid chromatography-mass spectrometry/mass spectrometry. This method offers a practical approach to studies of heterologous expression of anthocyanin biosynthesis pathway- related genes in tobacco.
